Naukowcy z Centre for Genomic Regulation postanowili opracować innowacyjną terapię, która byłaby w stanie zwalczyć bakterię Staphylococcus aureus, drobnoustrój, który najczęściej atakuje implanty medyczne – cewniki, rozruszniki serca czy protezy stawów. Bakteria jest niezwykle groźna, a pacjenci coraz częściej zakażają się nią w szpitalach. S. aureus wytwarza biofilm i jest oporny na niemal wszystkie antybiotyki, co znacznie utrudnia klinicystom skuteczne leczenie.

Bakterie często walczą ze sobą o terytorium, produkują i uwalniają enzymy przeciwko sobie. Hiszpańscy naukowcy postanowili wykorzystać ten proces na swoją korzyść. Pod lupę wzięto bakterię o nazwie Mycoplasma pneumoniae, którą zmodyfikowano w taki sposób, by nie była groźna dla człowieka i nie wywoływała infekcji. Bakteria wydajnie wytwarza dwa enzymy, które rozpuszczają biofilm i ściany komórkowe opornych na antybiotyki drobnoustrojów. M. pneumoniae posiada również niskie ryzyko mutacji i nie przenosi zmodyfikowanych genów na pobliskie drobnoustroje. Ponadto bakteria nie posiada ściany komórkowej, co pozwala jej na szybkie uwalnianie cząsteczek terapeutycznych zwalczających infekcje.

Skuteczność terapii przetestowano na modelu mysim. Kolonie M. pneumoniae wprowadzone zostały do cewników zainfekowanych S. aureus. Badania wykazały, że bakterie w 82 proc. skutecznie zwalczały infekcje u myszy.

Terapia „żywym lekiem” może znaleźć zastosowanie w leczeniu wielu infekcji

Prof. Luis Serrano, dyrektor CRG i współautor badania, podkreśla, że bakterie M. pneumoniae mogą przenosić dowolne białko terapeutyczne i leczyć źródło infekcji. Gdy dotrą do zainfekowanego miejsca w sposób ciągły produkują enzymy zwalczające oporne drobnoustroje. Ekspert dodaje, że  drobnoustroje M. pneumoniae można modyfikować i ukierunkowywać na różne choroby. Zdaniem prof. Serrano rozwiązanie to będzie miało wiele zastosowań w przyszłości.

María Lluch, współautorka badania dodaje, że technologia spełnia wszelkie standardy bezpieczeństwa i skuteczności. Informuje również, że pierwszym z celów nowej terapii są infekcje w układzie oddechowym. Naukowcy planują rozpoczęcie badań klinicznych w 2023 r.
